Component research

KLOW

KPV adds uptake and inflammatory-signalling questions to the individual components’ matrix, vascular and actin-related themes. The finished KLOW blend remains a separate research question.

Research summary

GHK-CU study

In vivo stimulation of connective tissue accumulation by the tripeptide-copper complex glycyl-L-histidyl-L-lysine-Cu2+ in rat experimental wounds.

The Journal of clinical investigation 1993

Maquart FX, Bellon G, Chaqour B, Wegrowski J, Patt LM, Trachy RE, Monboisse JC, Chastang F, Birembaut P, Gillery P

Rat wound-chamber model

GHK-Cu increased collagen and glycosaminoglycan accumulation in a rat wound-chamber model, with higher type I and III collagen messenger RNA. Dermatan sulfate also formed a larger share of the measured glycosaminoglycans, while TGF-β messenger RNA did not increase. The concentration-dependent changes show that matrix composition, not collagen alone, is relevant to the response in this artificial wound environment.

GHK-CU study

Effects of topical copper tripeptide complex on CO2 laser-resurfaced skin.

Archives of facial plastic surgery 2006

Leyden JJ, Stephens TJ, Herndon JH

Small randomised human post-laser study

A small human trial found no objective advantage for a GHK-Cu skin-care regimen after circumoral CO₂ laser resurfacing. Among the 13 people who completed it, computer analysis and blinded assessment found no significant difference in redness resolution, wrinkles or overall skin quality. Participants using GHK-Cu did report significantly greater improvement in their own assessment of overall skin quality at 12 weeks. The contrast between objective and subjective outcomes is useful, but the tiny sample offers only limited human context.

BPC-157 study

Gastric pentadecapeptide BPC 157 accelerates healing of transected rat Achilles tendon and in vitro stimulates tendocytes growth.

Journal of orthopaedic research 2003

Rat tendon and ligament injury models

BPC-157-treated rats had stronger functional and biomechanical measurements after Achilles tendon transection, together with more organised repair tissue than saline controls. In related cell experiments, BPC-157 opposed growth inhibition caused by the reactive aldehyde 4-hydroxynonenal. The tissue and cell results address different parts of an acute repair response and remain preclinical observations.

KPV study

PepT1-mediated tripeptide KPV uptake reduces intestinal inflammation.

Gastroenterology 2008

Dalmasso G, Charrier-Hisamuddin L, Nguyen HT, Yan Y, Sitaraman S, Merlin D

Intestinal cells and mouse colitis models

KPV entered intestinal epithelial and immune-cell preparations through PepT1, with lower NF-κB and MAP-kinase signalling and reduced inflammatory cytokine output. In two mouse colitis models, inflammatory tissue and cytokine measures also decreased. The work connects peptide transport with intracellular signalling, while keeping cell uptake and whole-tissue responses as separate measurements.

BPC-157 study

The promoting effect of pentadecapeptide BPC 157 on tendon healing involves tendon outgrowth, cell survival, and cell migration.

Journal of applied physiology (Bethesda, Md. 2011

Chang CH, Tsai WC, Lin MS, Hsu YH, Pang JH

Cellular and animal mechanistic research

BPC-157 increased tendon-fibroblast migration, spreading and survival under oxidative stress without directly increasing ordinary cell proliferation. FAK and paxillin phosphorylation rose, while total amounts of those proteins did not. The result connects cell movement with focal-adhesion signalling in rat-derived tendon cells.

TB-500 / full-length thymosin β4 study

Widely distributed residues in thymosin beta4 are critical for actin binding.

Biochemistry 2008

Au JK, Olivares AO, Henn A, Cao W, Safer D, De La Cruz EM

Molecular and analytical characterisation

Actin binding depended on residues distributed across full-length thymosin β4, not only its central short motif. Several substitutions markedly weakened association or stability of the actin complex. This helps explain why the complete molecule and an isolated fragment are distinct biochemical tools.

GHK-CU study

The tripeptide-copper complex glycyl-L-histidyl-L-lysine-Cu2+ stimulates matrix metalloproteinase-2 expression by fibroblast cultures.

Life sciences 2000

Siméon A, Emonard H, Hornebeck W, Maquart FX

Cellular and animal mechanistic research

GHK-Cu increased MMP-2 expression and secretion of its regulators TIMP-1 and TIMP-2 in dermal fibroblast cultures. Copper ions reproduced the MMP-2 response, whereas copper-free GHK did not. The finding broadens matrix biology from making connective tissue to regulating its breakdown and remodelling.

Models studied

  • GHK-CU extracellular-matrix models
  • BPC-157 tissue-response models
  • KPV intestinal-cell and mouse models
  • Unstudied four-component blend

Key findings

  • KPV uptake and inflammatory signals

PepT1-dependent uptake connects KPV with inflammatory-signalling measurements in cells and mouse colitis models.

  • BPC-157 cell migration

Cell movement and stress survival changed without a direct rise in ordinary proliferation; focal-adhesion phosphorylation helps distinguish those responses.

  • Full-length thymosin β4 actin binding

Contacts across the full thymosin β4 chain contribute to actin binding, making an isolated short motif a different experimental input.

  • GHK-Cu matrix turnover

MMP-2 and TIMP changes extend the research theme to turnover; the copper-only comparison helps separate peptide and metal contributions.
